Peptide Synthesis Resins

 

Solid Phase Peptide Synthesis (SPPS) was first introduced by Robert Bruce Merrifield, revolutionizing peptide synthesis by offering higher efficiency compared to liquid-phase synthesis. This method enables the rapid assembly of peptide chains through successive reactions of amino acid derivatives on an insoluble support, making it the preferred technique for synthesizing complex peptides.

Peptides can be obtained through biosynthesis (extracted from bacterial cultures such as Polymyxa or Pseudomonas aeruginosa) or chemical synthesis, which is categorized into:

  • Liquid-phase peptide synthesis (LPPS): Ideal for short peptide production

  • Solid-phase peptide synthesis (SPPS): Suitable for long-chain, cyclic, and complex peptides

SPPS is further classified into two main approaches:

  • Boc-based synthesis

  • Fmoc-based synthesis

By using a solid support, SPPS enables stepwise amino acid condensation, offering key advantages such as simplified purification, shorter synthesis cycles, and automation compatibility.
